The museum's roots lie in a municipal collection of antiquities that Ladenburg set up in 1904 in the recently acquired Stich'sches Haus; the Baden heritage conservator Ernst Wagner looked after the holdings. In 1909, the museum was officially opened in the presence of Grand Duke Friedrich II of Baden. After the interruption of World War II, it reopened in 1950 under archaeologist Berndmark Heukemes, who researched the town's Roman past extensively.

In 1968, the museum moved to the Bischofshof on Amtshof, the former residence of the Bishops of Worms. The building stands on the site of a medieval royal court and above remains of Roman Lopodunum; its core predates the 13th century. In the late 16th and early 17th centuries, it received Renaissance additions such as the octagonal stair tower with an onion-shaped cap (welsche Haube) and an oriel; Mannerist wall paintings came to light during restoration in the 1970s.

On four floors, the permanent exhibition spans Neolithic settlements, the Roman town of Lopodunum – capital of the Civitas Ulpia Sueborum Nicrensium – and the Migration Period, through the Middle Ages under the Bishops of Worms and the Electoral Palatinate administration, to modern Ladenburg. Separate sections are devoted to painted farmhouse furniture from the Odenwald and to the town's Jewish community; the exhibition "Neighbors 1938" on Ladenburg's Jews, created at the museum in 2018, was later also shown at Holocaust Museum Houston. Special exhibitions address current excavations and topics in the town's history.
